Unlocking epigenetic potential in polyploid wheat – new study out now
Knocking out DNA methyltransferase genes usually kills diploid crops, which has kept breeders from tapping into epigenetic diversity. But polyploidy offers a clever way around that.
In a fresh paper from the *Journal of Experimental Botany*, researchers used wheat’s polyploid “cushion” to create CRISPR-edited alleles of *MET1-1* in tetraploid and hexaploid lines. By tracking seed genotype ratios and combining whole‑genome bisulfite sequencing (WGBS) with RNA‑seq, they found:
✅ Viable plants carrying up to four mutant copies
✅ A stepwise, dosage‑dependent loss of CG methylation
✅ Heritable changes in flowering time and awn length – not linked to the targeted locus
This shows that polyploidy can “titrate” epigenetic strength without causing lethality, opening a non‑transgenic route to fine‑tune complex agronomic traits.
